We are visiting WDW in May with our two boys, 3 and 6, but only for one day. This will be our first experience and I have a million questions. To get the most out of the day, should we get park hopper tickets or stick with one park? Any tips?

I think to get the most out of one day, I'd go to one park and do it really well.  There is so much to see and enjoy at each park, you will have plenty to do in one day.  With the ages of your  children, I think the Magic Kingdom would be a good fit for your family and it always seems to epitomize the Disney experience.  You could always work in a break during the day or consider having a meal by visiting some of the nearby resorts that are on the monorail:  The Polynesian, The Contemporary, and the Grand Floridian.  Your sons may even find being on the monorail another ride!
If you aren't staying at a WDW resort, I'd try and go to the park on a day that does not have an Extra Magic Hour for resort guests.  That way you can arrive without there being a crowd already in the park.  Here is a link to the Extra Magic Hours schedule to help with your planning.
Also, make sure to utilize fast-passes to avoid waiting in lines as much as possible to make the most out of your day.  Have a magical trip!
