I'm going to WDW in April with my sisters, 5yo nephew and 1yo niece. We'll be brining a stroller for the 1yo but does anyone think we'll need one for the 5yo too?

That's a tough question, Jennifer.  It all depends upon the five-year-old in question.  Our most recent trip was in October of last year.  My son was just over five-years-old at the time and we did, indeed, bring a stroller for him.  Here's the catch, though: we didn't end up using it a single time.  We know from experience how much walking we do during our Disney trips and we thought for sure our little guy would tucker out and end up in the stroller.  On our first park day, we opted to leave it behind and we found he did just fine without it.  So we left it in our resort room again on the second day and, again, he was fine.  By the end of our vacation we honestly hadn't used that stroller a single time, so it proved to be just another thing to lug home.  I wish we hadn't brought it!

All that said, your experience may vary because your nephew might not be up for all that walking.  My {seasoned} advice would be to go without a stroller for him.  Do, though, Google the term "Orlando stroller rental" before you leave home and jot down the phone numbers for a couple of the companies.  That way, if worse comes to worse, you could always call and have a stroller delivered (subject to availability) during your trip.  A word of warning, though, you will do a LOT of walking during your vacation.  Walking that can be measured by miles, not feet.  So if that seems like too much for your nephew, then go ahead and bring along a stroller.  Good luck!
