My family will arrive on June 27. I plan to purchase a 7 day park hopper +waterpark. If we go to Disney Quest on our arrival date, will this also take away one of Park days? I wasn't sure if by going to a waterpark or extra first will activate the tickets

Hi, Marilyn! The Theme Park tickets and Water Parks Fun and More options are counted separately.  When you go to DisneyQuest (or any of the Water Parks Fun and More options), it subtracts one of those option entitlements. It does not impact your Theme Park tickets at all. So after spending your first day at DisneyQuest, you will still have seven Theme Park days and six more Water Parks Fun and More options left. Theme Park days are only subtracted when you enter the Magic Kingdom, Epcot, Disney's Hollywood Studios, or Animal Kingdom. Happy planning and have a Magical vacation!
